I recommend that you find the absolute smallest pack possible. If it's only going to carry a day's clothes in it, then I believe you can do that in a reasonably small pack.
The Museums generally have a limit as to how big of a pack you can take in with you. But, with a small pack, you should be just fine. Think in terms of the kinds of packs that high school students probably use these days for carrying books.
I like to travel light myself. I've actually gone on business trips and only packed one extra shirt. I would simply wash my other shirt in the hotel sink using ivory soap and then let it dry near the heating vents.
If all else fails, I THINK Union Station (the train station in Wash DC) has a service where they will store bags for a few hours. It might cost like $5 to $8 though.

You'll be fine. Like everyone else has said, they'll probably go through your bags at the museums but that's about it. And seriously, bring water. And an umbrella, we've been having a lot of rain. I hope you have fun while you're here!

If you take it into a Smithsonian museum, they very well may search it. If you're ok with that, then there is no problem.

Agreed with the person above. DC is a big city. It is VERY walkable to, therefor people walk everywhere. People take backpacks everywhere they go. It's not a big deal, whatsoever. Take your backpack. Bring a waterbottle too. DC is HOT! Enjoy it here!

No you won't have an issue carrying your backpack around. They're the norm around here, I carry mine everywhere I go. If a place ie museum has a problem with you bringing in the bag they usually have an area where you can check it for little to no cost.
